Overview

Autologous MAGE-C2-specific TCR T cells is an investigational adoptive cell therapy developed by Erasmus University Medical Center (Erasmus MC). The therapy consists of a patient's own T cells that have been genetically modified to express a T-cell receptor (TCR) targeting the MAGE-C2 (MC2) cancer-germline antigen, specifically the ALK epitope presented by HLA-A2. MAGE-C2 is a tumor-selective antigen highly expressed in melanoma and head and neck squamous cell carcinoma (HNSCC) but absent in healthy adult tissues. The treatment protocol involves the administration of these engineered T cells following epigenetic modulation with azacitidine and valproic acid to enhance antigen expression, alongside low-dose interleukin-2 (IL-2) to support T-cell expansion and persistence. It is currently being evaluated in a Phase I/II clinical trial for patients with advanced melanoma or recurrent/metastatic HNSCC.
