This autologous mature dendritic cell vaccine is a personalized cellular immunotherapy developed by John Kirkwood and colleagues at the University of Pittsburgh. The vaccine is produced by harvesting a patient's own peripheral blood mononuclear cells, which are then differentiated into dendritic cells and matured ex vivo using a specific cytokine cocktail (typically including IL-1b, IL-6, TNFa, and PGE2). These mature dendritic cells are pulsed with autologous tumor lysate, which provides a comprehensive array of patient-specific tumor-associated antigens. When re-administered to the patient via ultrasound-guided intranodal injection, these professional antigen-presenting cells prime and activate tumor-specific T lymphocytes, aiming to induce a systemic anti-tumor immune response against metastatic melanoma.
