This combination therapy consists of two distinct autologous cancer vaccines: an **autologous melanoma cell vaccine** (TCV), made by irradiating short-term cultured tumor cells derived from a melanoma patient's own metastasis, and an **autologous dendritic cell vaccine** (DCV), made by loading dendritic cells harvested from the patient ex vivo with tumor antigens derived from their own melanoma tissue[1][2]. Both vaccine platforms are intended to stimulate the patient's immune system to recognize and attack melanoma cells by presenting patient-specific tumor-associated antigens. The DCV, in particular, aims to enhance antitumor immunity by precisely delivering tumor antigens to dendritic cells, resulting in amplified CD8+ T cell responses and upregulated TH1/TH17 immunity, while the TCV provides direct antigen exposure from irradiated tumor cells[1][2]. The combination is used in the setting of metastatic or advanced melanoma and is considered a personal immunotherapy. Development has been led mainly by academic centers and biotechnology companies specializing in cellular immunotherapies[1][2].
