Autologous melanoma dendritic cell vaccine is a personalized cancer immunotherapy consisting of a patient's own dendritic cells (DCs) that have been matured and loaded ex vivo with autologous tumor antigens. The process typically involves harvesting monocytes from the patient's peripheral blood via leukapheresis, differentiating them into mature dendritic cells in culture, and pulsing them with tumor-associated antigens derived from the patient's own irradiated melanoma cells or tumor cell lysates. Once re-administered to the patient, these professional antigen-presenting cells migrate to secondary lymphoid organs where they present tumor antigens to T cells, thereby priming and activating a tumor-specific immune response. This therapeutic approach is primarily investigated for patients with advanced metastatic melanoma (Stage III or IV) to improve survival outcomes by leveraging the patient's own immune system to target and eliminate residual or metastatic cancer cells.
