The **autologous melanoma vaccine** is a personalized cancer immunotherapy created using tumor cells derived from an individual patient's own melanoma. The vaccine typically consists of autologous melanoma cells harvested from the patient's surgical specimen or from cultured tumor cell lines. These cells may be conjugated with immunostimulatory haptens (such as dinitrophenyl), mixed with an adjuvant (often Bacillus Calmette-Guerin, BCG), and irradiated to inactivate them. The vaccine can also be formulated with immune stimulators such as granulocyte-macrophage colony-stimulating factor (GM-CSF), or used to load autologous dendritic cells with tumor antigens before injection. The mechanism of action centers on the induction of a patient-specific anti-melanoma immune response, particularly the activation of T cells against melanoma-associated antigens, which enhances immune surveillance and tumor cell eradication. The approach has shown promising results in phase I/II trials, especially as adjuvant therapy for high-risk, surgically resected melanoma and metastatic disease[1][2][3][5][6][9].
