Overview

Autologous mesothelin-specific TCR-T cells refer to a class of adoptive cellular therapies where a patient's own T cells are genetically engineered to express a T-cell receptor (TCR) targeting mesothelin (MSLN). Mesothelin is a cell-surface glycoprotein overexpressed in various solid tumors, including pancreatic ductal adenocarcinoma, mesothelioma, and ovarian cancer, while maintaining limited expression in normal tissues. A prominent example of this therapy is FH-TCR-Tmsln, developed by the Fred Hutchinson Cancer Center. This specific construct uses a high-affinity TCR to recognize mesothelin peptides in the context of HLA-A*02:01. The treatment typically involves lymphodepleting chemotherapy (e.g., cyclophosphamide and fludarabine) followed by the infusion of the engineered T cells, which then expand and target mesothelin-positive malignant cells.
