Autologous monocyte-derived dendritic cells pulsed with autologous tumor lysate is an experimental, personalized cell-based immunotherapy developed by Erasmus Medical Center Rotterdam for the treatment of malignant pleural mesothelioma. The therapy involves harvesting a patient's own monocytes, differentiating them into dendritic cells ex vivo, and loading them with a lysate derived from the patient's own tumor tissue. This process allows the dendritic cells to present a broad spectrum of patient-specific tumor antigens to the immune system. Upon re-administration, these cells aim to activate and expand tumor-reactive CD4+ and CD8+ T cells. In clinical studies (NCT00280982), the vaccine was administered alongside keyhole limpet hemocyanin (KLH) as an immunological marker following standard chemotherapy.
