Autologous monocyte-derived dendritic cells transfected with DNA are a form of personalized cell therapy in which a patient's own (autologous) monocytes are differentiated into dendritic cells ex vivo and then genetically modified by introducing plasmid DNA encoding tumor-associated antigens or immunomodulatory molecules. These engineered dendritic cells are designed to present the encoded antigens to T lymphocytes, thereby stimulating antigen-specific cytotoxic T lymphocyte (CTL) responses against cancer. This approach is being investigated primarily as an immunotherapy for melanoma and other cancers, leveraging the ability of gene-modified DCs to induce broad cellular immune responses without requiring prior knowledge of patient MHC haplotypes or specific peptide epitopes. The process can be further enhanced by co-transfecting cytokine genes such as IL-12 or IFN-alpha to bias immune responses toward Th1-type immunity[1][2][5].
