This investigational combination immunotherapy regimen is being developed by Universitair Ziekenhuis Brussel for the treatment of recurrent glioblastoma. The regimen, evaluated in the GlitIpNi clinical trial, involves the per-operative intratumoral administration of autologous CD1c(BDCA-1)+ and CD141(BDCA-3)+ myeloid dendritic cells (myDC) into the brain tissue following neurosurgical resection. This is combined with the localized administration of the CTLA-4 inhibitor ipilimumab and the PD-1 inhibitor nivolumab into the resection cavity, followed by repeated intracavitary and systemic intravenous administrations of nivolumab. The mechanism of action relies on the synergy between dendritic cell-mediated antigen presentation and dual checkpoint blockade to overcome the immunosuppressive microenvironment of the central nervous system and stimulate a robust anti-tumor T-cell response.
