Overview

Autologous nano CD5-CAR T cells are an investigational chimeric antigen receptor (CAR) T-cell therapy designed to treat relapsed or refractory T-cell malignancies, specifically T-cell acute lymphoblastic leukemia (T-ALL) and T-cell lymphoblastic lymphoma (T-LBL). The therapy utilizes a patient's own T cells, which are genetically modified to express a CAR targeting the CD5 antigen, a glycoprotein found on the surface of most mature T cells and T-cell leukemias. A key innovation in this therapy is the use of a nanobody-based (VHH) binding domain instead of a conventional single-chain variable fragment (scFv), which may enhance the stability and specificity of the CAR. Developed by the Institute of Hematology & Blood Diseases Hospital in China, the therapy is currently undergoing Phase 1/2 clinical evaluation to assess its safety, optimal dosing, and efficacy in patients with advanced T-cell cancers.
