Autologous natural killer T (NKT) cell therapy is an adoptive cell transfer biologic developed by the Peking University Cancer Hospital & Institute for the treatment of advanced melanoma. The therapy involves the ex vivo isolation and expansion of CD8+ NKT cells from a patient's peripheral blood mononuclear cells (PBMCs). This expansion is achieved using a specific cocktail of cytokines, including Interleukin-2 (IL-2), Interleukin-4 (IL-4), and Granulocyte-Macrophage Colony-Stimulating Factor (GM-CSF), along with the anti-CD3 monoclonal antibody OKT3. The resulting cell product consists of autologous CD8+ NKT cells, a unique lymphocyte population that expresses both T-cell receptors and natural killer cell markers. These cells are intravenously re-infused into the patient to exert anti-tumor effects through direct cytotoxicity and immune system modulation. A Phase I clinical trial (NCT02619058) demonstrated that the therapy is well-tolerated and shows preliminary efficacy in patients with metastatic melanoma.
