Overview

Autologous natural killer T (NKT) cells are an adoptive cell therapy being developed by the Chinese PLA General Hospital for the treatment of various advanced, treatment-refractory solid tumors. NKT cells represent a unique lineage of T cells that express both a semi-invariant T-cell receptor (TCR) and natural killer (NK) cell markers. These cells are characterized by their ability to recognize glycolipid antigens presented by the non-polymorphic MHC class I-like molecule CD1d. In this therapeutic approach, NKT cells are isolated from the patient's peripheral blood, expanded and activated ex vivo, and subsequently re-infused. Upon re-infusion, these cells exert anti-tumor effects through direct cytotoxicity and the rapid production of large quantities of Th1-type cytokines, such as interferon-gamma (IFN-γ), which further orchestrate and enhance the host's overall anti-tumor immune response by activating NK cells and CD8+ T cells.
