Overview

YS247 is an autologous neoantigen-pulsed dendritic cell vaccine being developed by Changhai Hospital for the treatment of metastatic castration-resistant prostate cancer (mCRPC). This personalized immunotherapy involves isolating a patient's peripheral blood mononuclear cells (PBMCs) and differentiating them into dendritic cells (DCs) ex vivo using GM-CSF and IL-4. These DCs are then pulsed with synthetic neoantigen peptides, which are identified through next-generation sequencing (NGS) of the patient's specific tumor tissue to capture unique mutations. Following maturation with a cytokine cocktail (TNF-α, IFN-γ, and PGE2), the vaccine is administered subcutaneously. The goal is to leverage the matured DCs to present these neoantigens to the patient's T cells, thereby inducing a targeted and robust anti-tumor immune response.
