This investigational cell therapy consists of autologous natural killer (NK) cells expanded ex vivo for the treatment of advanced malignancies. Developed and evaluated at Hangzhou Cancer Hospital, the therapy involves isolating NK cells from a patient's own peripheral blood, stimulating their proliferation and activation in a laboratory setting, and reinfusing them into the patient. In the specific clinical context of Hangzhou Cancer Hospital's Phase I research, these cells are evaluated in combination with nimotuzumab, a humanized monoclonal antibody targeting the epidermal growth factor receptor (EGFR). The therapeutic strategy leverages the NK cells' expression of Fc receptors (CD16), which bind to the Fc portion of nimotuzumab-coated tumor cells to trigger antibody-dependent cellular cytotoxicity (ADCC). Additionally, the transferred NK cells provide direct, non-specific anti-tumor activity through the release of cytotoxic granules containing perforin and granzymes.
