Overview

Autologous NY-ESO-1-specific CD4+ T cells are an adoptive cell therapy involving the isolation, ex vivo expansion, and re-infusion of a patient's own helper T lymphocytes that have been sensitized to the cancer-testis antigen NY-ESO-1 (New York esophageal squamous cell carcinoma 1). Unlike many T-cell therapies that focus on cytotoxic CD8+ T cells, this approach leverages the multifaceted role of CD4+ T cells, which can provide help to CD8+ cells, induce inflammatory cytokines like IFN-gamma, and in some cases, exhibit direct MHC class II-restricted cytotoxicity against tumor cells. The therapy is typically investigated for NY-ESO-1-expressing solid tumors, such as metastatic melanoma and synovial sarcoma. It often involves the use of lymphodepleting chemotherapy prior to infusion to enhance cell persistence and may be administered alongside interleukin-2 (IL-2) to support T-cell survival and expansion in vivo.
