Overview

Autologous NY-ESO-1-specific CD8-positive T lymphocytes are a form of adoptive cell therapy in which a patient's own (autologous) CD8+ T cells are isolated and engineered or selected to specifically recognize the cancer-testis antigen NY-ESO-1. This antigen is highly immunogenic and expressed in various tumors, including melanoma, synovial sarcoma, ovarian carcinoma, fallopian tube carcinoma, myxoid liposarcoma, and others. The therapy involves expanding these specific cytotoxic T cells ex vivo before reinfusing them into the patient to target and destroy tumor cells expressing NY-ESO-1. The mechanism of action is direct recognition and killing of tumor cells presenting NY-ESO-1 peptides via HLA class I molecules on their surface. This approach has shown clinical responses in early-phase trials for advanced cancers refractory to standard treatments[3][7][10].
