Overview

Autologous T cells encoding a chimeric antigen receptor (CAR) targeting human B-cell maturation antigen (BCMA) represent a class of personalized cell therapies primarily used to treat multiple myeloma. The process involves collecting a patient's T cells, genetically modifying them (often using a lentiviral or retroviral vector) to express a CAR specific to BCMA, and then expanding and re-infusing them into the patient. BCMA is a cell surface receptor highly and selectively expressed on mature B lymphocytes and plasma cells, making it an ideal target for multiple myeloma. Once infused, these CAR-T cells recognize and eliminate BCMA-positive malignant cells. Approved therapies in this category include idecabtagene vicleucel (Abecma) and ciltacabtagene autoleucel (Carvykti), while others like Novartis's PHE885 are in clinical development.
