The autologous TLPLDC (Tumor Lysate, Particle-Loaded, Dendritic Cell) vaccine is a personalized, cell-based cancer immunotherapy developed by Perseus PCI and Elios Therapeutics. It is a biologic/cellular therapy with no small-molecule active ingredient; instead, its active components are autologous tumor lysate (TL) loaded into yeast cell wall particles (YCWP), which are then phagocytosed ex vivo by autologous monocyte-derived dendritic cells (DCs) harvested from the patient's own blood. The resulting vaccine is injected intradermally to stimulate a patient-specific anti-tumor immune response. The biological target is the patient's own tumor-associated neoantigens, presented via dendritic cells to activate the immune system. It has been studied in phase I/IIa and phase IIb clinical trials for solid tumors broadly, with a particular focus on resected stage III/IV melanoma, as well as other solid tumors in the adjuvant and metastatic settings.
