The **autologous total tumor RNA-loaded monocyte-derived mature dendritic cell vaccine** is an investigational, patient-specific cellular immunotherapy developed by the Academisch Medisch Centrum (AMC, Amsterdam UMC). Classified as an Advanced Therapy Medicinal Product (ATMP), this personalized vaccine is manufactured by extracting peripheral blood monocytes from a patient via leukapheresis, differentiating them ex vivo into mature dendritic cells, and loading them with the full repertoire of tumor-associated antigens via electroporation or pulsing with total tumor RNA extracted from the patient's own tumor tissue. The vaccine is designed to activate tumor-antigen-specific cytotoxic T lymphocytes (CTLs), thereby inducing a robust anti-tumor immune response tailored to the patient's unique tumor profile. It has been primarily studied in manufacturing-validation clinical trials for the treatment of esophageal adenocarcinoma and pancreatic cancer.
