This combination immunotherapy protocol, developed by the H. Lee Moffitt Cancer Center and Research Institute, is designed for the treatment of mantle cell lymphoma (MCL). The regimen involves an autologous tumor cell vaccine formulated with a universal bystander cell line (GM.CD40L) that has been genetically engineered to secrete Granulocyte-Macrophage Colony-Stimulating Factor (GM-CSF) and express CD40 Ligand (CD40L). This vaccine is administered alongside low-dose interleukin-2 (IL-2). The mechanism of action relies on the GM.CD40L cells to activate host dendritic cells and enhance the presentation of tumor antigens derived from the patient's own harvested MCL cells. Simultaneously, IL-2 acts as a cytokine support to promote the expansion and effector function of tumor-specific T-cells. This therapeutic approach is typically employed as a consolidation strategy following induction chemotherapy to eradicate residual disease and prolong remission in patients with MCL.
