Overview

The autologous tumor DNA-transfected fibroblast vaccine is an investigational personalized cancer immunotherapy. It consists of a patient's own fibroblasts that have been genetically modified (transfected) with DNA extracted from the patient's own tumor cells. The rationale behind this approach is to use the fibroblasts as vehicles to express a broad spectrum of tumor-associated antigens (TAAs) unique to the individual's malignancy. Upon administration, these modified fibroblasts are intended to act as antigen-presenting cells or to facilitate the cross-presentation of TAAs to the host immune system, thereby stimulating a robust cytotoxic T-lymphocyte (CTL) response against tumor cells expressing the same antigens. This strategy aims to overcome the limitations of single-antigen vaccines by targeting the full repertoire of a patient's specific tumor mutations.
