The autologous tumor-loaded alpha-type 1 dendritic cell (αDC1) vaccine is a personalized cancer immunotherapy developed by the Roswell Park Comprehensive Cancer Center. This cell-based vaccine is produced by isolating a patient's own monocytes and differentiating them into alpha-type-1 polarized dendritic cells, which are then loaded with autologous tumor lysate derived from the patient's surgically resected tumor tissue. The αDC1 cells are matured using a specific polarizing cocktail (typically including IL-1β, TNF-α, IFN-α, IFN-γ, and poly-L-lysine) to enhance their ability to produce high levels of IL-12p70 and stimulate potent Th1-type anti-tumor T-cell responses. Administered via intranodal or intradermal injection, the vaccine is primarily investigated as an adjuvant therapy following cytoreductive surgery for peritoneal surface malignancies, including metastatic pancreatic cancer and malignant peritoneal mesothelioma, to target residual microscopic disease and prevent recurrence.
