Autologous tumor lysate is a personalized biological product used as an experimental immunotherapy for the treatment of sarcoma. Prepared from a patient's own surgically resected tumor tissue, the lysate contains a comprehensive array of tumor-associated antigens (TAAs) unique to that individual's malignancy. In the clinical trial context led by Macarena De La Fuente, MD (NCT01803152), the lysate is used both to load autologous dendritic cells for a primary vaccine and as a standalone intradermal booster. The mechanism of action relies on the presentation of these antigens to the immune system, particularly T-cells, to stimulate a targeted attack against residual or metastatic cancer cells. This approach is designed to bypass the immune evasion strategies of sarcomas by providing a diverse set of targets for immune recognition.
