The autologous tumor lysate-loaded cross-presenting dendritic cell vaccine is an investigational personalized cell therapy developed by Stichting Radboud universitair medisch centrum for the treatment of ovarian cancer. This vaccine utilizes naturally occurring cross-presenting dendritic cells, specifically the cDC1 (BDCA3+) subset, which are isolated directly from the patient's peripheral blood. Unlike traditional monocyte-derived dendritic cell vaccines, these natural DCs possess superior innate capabilities for cross-presentation. The cells are loaded ex vivo with autologous tumor lysate, providing a comprehensive array of patient-specific tumor antigens. Upon re-administration via intranodal injection, these dendritic cells present tumor-derived peptides on MHC class I and MHC class II molecules to activate cytotoxic CD8+ T cells and helper CD4+ T cells, respectively. This dual activation is designed to overcome tumor-induced immunosuppression and generate a targeted, systemic anti-tumor immune response.
