Overview

This autologous dendritic cell-based vaccine is a personalized cellular immunotherapy developed at Dartmouth-Hitchcock Medical Center for the treatment of metastatic renal cell carcinoma (mRCC). The vaccine is produced by harvesting peripheral blood monocytes from the patient, which are then differentiated into dendritic cells ex vivo and loaded with autologous tumor lysate. This loading process allows the dendritic cells to present a comprehensive and individualized array of tumor-associated antigens specific to the patient's own malignancy. Administered via intranodal injection, the vaccine aims to prime and activate the patient's T-cells to recognize and destroy tumor cells. In clinical development, this vaccine has been studied in combination with systemic immunotherapies such as aldesleukin (IL-2) and interferon-alpha-2b, as well as the anti-VEGF monoclonal antibody bevacizumab, to enhance the overall anti-tumor immune response.
