This combination immunotherapy regimen, developed by Wuerzburg University Hospital, is designed for the treatment of relapsed high-grade gliomas in children and adolescents. The protocol involves an individualized cancer vaccine using autologous, tumor-lysate loaded dendritic cells to prime the immune system against tumor antigens. To optimize this response, metronomic cyclophosphamide is administered to deplete immunosuppressive regulatory T cells (Tregs). This is followed by dual checkpoint blockade with nivolumab (targeting PD-1) and ipilimumab (targeting CTLA-4) to prevent T-cell exhaustion and enhance the magnitude and duration of the anti-tumor immune response.
