This autologous dendritic cell vaccine is an investigational cell-based immunotherapy developed by Heinrich-Heine-Universitaet Duesseldorf for the treatment of glioblastoma. The manufacturing process involves isolating monocytes from the patient's peripheral blood, which are then differentiated into mature dendritic cells (DCs) using a specific cytokine maturation cocktail (typically including TNF-alpha, IL-1beta, IL-6, and PGE2). These mature DCs are pulsed with a lysate derived from the patient's own resected tumor tissue, allowing the cells to process and present a broad spectrum of patient-specific tumor-associated antigens. Once administered, these professional antigen-presenting cells migrate to the lymph nodes where they activate CD8+ cytotoxic T lymphocytes and CD4+ helper T cells, inducing a targeted and polyvalent immune response against the heterogeneous cell populations within the glioblastoma.
