Overview

This is an autologous dendritic cell-based cancer vaccine developed at Cedars-Sinai Medical Center for the treatment of malignant gliomas, including glioblastoma multiforme and anaplastic astrocytoma. The vaccine is a personalized biologic immunotherapy created by harvesting a patient's own monocytes, differentiating them into dendritic cells ex vivo, and pulsing them with a lysate derived from the patient's own surgically resected tumor tissue. This process loads the dendritic cells with a broad spectrum of tumor-associated antigens unique to the individual's cancer. Once re-administered to the patient, these mature dendritic cells present the antigens to the immune system, specifically activating cytotoxic T lymphocytes (CTLs) to recognize and destroy residual glioma cells. In clinical trials, this vaccine has often been evaluated in combination with topical imiquimod, a toll-like receptor 7 (TLR7) agonist, to enhance the anti-tumor immune response.
