The autologous tumor-pulsed dendritic cell vaccine developed by the University of Michigan Rogel Cancer Center is an individualized cellular immunotherapy investigated for the treatment of metastatic colorectal cancer. The therapeutic process involves harvesting a patient's own dendritic cells (DCs) from peripheral blood, which are then matured ex vivo and pulsed with antigens derived from the patient's own autologous tumor lysate. Upon re-administration, these dendritic cells act as professional antigen-presenting cells to prime and activate the patient's T cells against tumor-specific antigens. In clinical trial contexts (such as NCT00004149), this vaccine was administered in combination with subcutaneous Interleukin-2 (IL-2) to further stimulate the expansion and effector function of the induced tumor-reactive cytotoxic T lymphocytes.
