The autologous tumor RNA-pulsed dendritic cell vaccine is an experimental, personalized cancer immunotherapy being developed by Academisch Medisch Centrum (AMC) for the treatment of inoperable pancreatic adenocarcinoma. This biologic therapy involves extracting RNA from a patient's own tumor tissue—specifically via fine-needle aspirates—and using it to pulse autologous dendritic cells ex vivo. The resulting vaccine presents the full repertoire of the individual's tumor-associated antigens to the immune system, aiming to elicit a robust cytotoxic T-cell response against the pancreatic cancer. Currently in the protocol optimization phase, the development focuses on a GMP-compatible linear amplification method to generate sufficient RNA for multiple vaccine doses from minimal tissue samples.
