Autologous tumour homogenate-loaded dendritic cells is an experimental personalized cancer immunotherapy developed by the Istituto Romagnolo Per Lo Studio Dei Tumori (IRST). This cell-based vaccine is produced by isolating a patient's own monocytes from peripheral blood, differentiating them into mature dendritic cells ex vivo, and then loading (pulsing) them with a homogenate derived from the patient's own surgically resected tumor tissue. This approach allows the dendritic cells to capture and present a comprehensive, polyvalent array of patient-specific tumor-associated antigens (TAAs) and neoantigens. Upon intradermal re-administration, these activated dendritic cells migrate to regional lymph nodes where they present the processed antigens to naive T cells. This process is designed to prime and expand a robust, tumor-specific immune response involving both cytotoxic CD8+ T lymphocytes and helper CD4+ T cells, specifically targeting the heterogeneous cell population of the patient's colorectal cancer.
