AUY922 + trastuzumab is a combination therapy that pairs the HSP90 inhibitor AUY922 with the HER2-targeting monoclonal antibody trastuzumab. AUY922 works by inhibiting heat shock protein 90 (HSP90), which enables the activation of many client proteins including HER2. This inhibition leads to degradation of oncogenic proteins. The combination has shown synergistic activity in HER2-positive cancer models, particularly in trastuzumab-resistant settings. The rationale for this combination is based on the ability of HSP90 inhibition to overcome trastuzumab resistance mechanisms in HER2-positive cancers.
