AV-1959D is a DNA-based epitope vaccine designed to induce an immune response against amyloid-beta (Aβ) for the prevention and treatment of Alzheimer's disease. The vaccine encodes three copies of the Aβ1–11 B cell epitope fused to the MultiTEP platform, which contains multiple universal foreign T helper epitopes recognized by diverse human MHC class II alleles. This design aims to stimulate robust anti-Aβ antibody production by activating both naïve and memory T helper cells while avoiding activation of potentially harmful autoreactive T cells. Delivered via electroporation, AV-1959D has demonstrated immunogenicity and safety in preclinical models (mice, rabbits, non-human primates), with no significant adverse neuropathological effects observed. The vaccine is currently being evaluated in Phase I clinical trials for early Alzheimer's disease[1][5][6][7].
