AV-1980R is an investigational **protein-based therapeutic vaccine** targeting pathological forms of the microtubule-associated protein tau, designed to prevent or treat Alzheimer’s disease. The vaccine is built on the MultiTEP platform and comprises an N-terminal peptide from tau’s phosphatase activating domain (PAD; residues 2–18), fused with highly immunogenic T-helper cell epitopes from foreign proteins. Its mechanism is to induce a strong and specific humoral immune response generating antibodies against the PAD of tau, which is implicated in tau aggregation and toxicity in Alzheimer’s disease. Unlike native tau, only pathological aggregated tau exposes the PAD, enabling the vaccine to target disease-associated tau with minimal off-target effects. Preclinical studies suggest the vaccine produces anti-tau antibodies recognizing neurofibrillary tangles and dystrophic neurites, and may reduce tau aggregation, glial activation, and associated cognitive impairment. The vaccine is in early human clinical testing for preclinical Alzheimer’s disease.[1][2][3][4][5][6][7]
