Overview

AV-203 is a humanized IgG1 monoclonal antibody that selectively targets and inhibits the receptor tyrosine-protein kinase erbB-3 (also known as HER3 or ERBB3). By blocking both ligand-dependent and ligand-independent ERBB3 signaling, AV-203 aims to disrupt oncogenic pathways involved in tumor growth and survival. The drug has demonstrated preclinical activity across multiple solid tumor models including breast, head and neck, lung, ovarian, and pancreatic cancers. Developed by AVEO Oncology (a subsidiary of LG Chem), AV-203 has completed Phase 1 clinical trials in patients with advanced solid tumors. In these studies it was generally well tolerated; one patient with squamous non–small-cell lung cancer expressing high levels of neuregulin 1 achieved a partial response. The recommended Phase 2 dose is 20 mg/kg IV every two weeks[1][2][4][5].
