Overview

AV-353 is a potent, selective, high-affinity inhibitory monoclonal antibody targeting the Notch 3 receptor pathway[1][2][4][16]. The Notch 3 signaling pathway plays a crucial role in cell-to-cell communication and gene regulation, affecting differentiation processes throughout the life cycle. AV-353 is being investigated primarily for its ability to antagonize Notch 3, which has been implicated in diseases such as pulmonary arterial hypertension (PAH), cancer (including triple-negative breast cancer and multiple myeloma), cardiovascular diseases, and neurodegenerative disorders[1][2][4][16]. Preclinical data suggest AV-353 could potentially reverse the phenotype of PAH, representing a disease-modifying approach rather than merely symptomatic relief[2][4][6]. The antibody’s selectivity for Notch 3 distinguishes it from pan-notch or ligand-inhibiting strategies.
