AV-380 + chemotherapy

AV-380 + chemotherapy is a combination of the investigational drug AV-380 (also known as rilogrotug)—a first-in-class, humanized inhibitory IgG1 monoclonal antibody targeting growth differentiation factor 15 (GDF-15)—administered together with standard of care chemotherapy regimens in patients with metastatic cancer and cancer-associated cachexia. AV-380 acts by inhibiting GDF-15, a pro-inflammatory cytokine implicated in the pathogenesis of cancer cachexia—a syndrome marked by severe involuntary weight loss, muscle wasting, and functional decline. In preclinical models, AV-380 reversed weight loss, restored muscle mass, and improved metabolic function by shifting from a catabolic to an anabolic state. The combination is currently being investigated in Phase 1b trials to assess safety, pharmacokinetics, pharmacodynamics, and potential additive clinical benefits in cachectic cancer patients on chemotherapy.
