AVA004

AVA004 is an Affimer-based protein therapeutic targeting programmed death-ligand 1 (PD-L1), developed by Avacta as an immune checkpoint inhibitor for the treatment of solid tumours. It is a small, engineered affinity protein formatted with an Fc domain for half-life extension, designed to block the PD-1/PD-L1 interaction and thereby restore T‑cell–mediated antitumour immunity, with potential advantages over monoclonal antibodies including smaller size, improved tissue penetration, and ease of manufacturing.[1][4][9][11][14][15] AVA004 has demonstrated potent PD-L1 antagonism and preclinical efficacy in syngeneic and xenograft mouse models, where it compares favourably with approved PD-L1 antibodies such as durvalumab and avelumab, and is intended as a foundational asset for future Affimer-based bispecifics, fusion proteins, and drug conjugates.[1][4][7][9][13][15]
