Drug intelligence / Profile preview
AVA032 is a preclinical-stage tumor-targeted immunocytokine comprising an anti-PD-L1 Affimer® scaffold protein fused to human interleukin-15 (IL-15) and the IL-15 receptor alpha (IL-15Rα) sushi domain. Developed by Avacta, the bispecific fusion protein is designed to target the tumor microenvironment by binding to programmed death-ligand 1 (PD-L1) on tumor cells, macrophages, and dendritic cells. This targeting delivers the immunostimulatory IL-15 cytokine directly to the tumor, where it activates the IL-2/IL-15 receptor beta/gamma (IL-2Rβ/γ) complex on CD8+ T cells and natural killer (NK) cells. By combining immune checkpoint inhibition (blocking the PD-1:PD-L1 pathway) with localized cytokine-mediated immune activation, AVA032 aims to reverse T-cell exhaustion and enhance anti-tumor immune responses while minimizing systemic cytokine toxicity.
