AVA04 + VbP is an investigational **tumor microenvironment activated drug conjugate** being developed by **Avacta** for cancer treatment. The construct combines **AVA04**, an Affimer-based **PD-L1 checkpoint inhibitor**, with **VbP**, an **I-DASH chemotherapy warhead**, linked through a proprietary **FAPα-cleavable** linker designed to release the cytotoxic payload preferentially in the tumor microenvironment rather than systemically. This design is intended to preserve PD-L1-directed antitumor immune activity while mitigating the acute systemic toxicity associated with I-DASH inhibitors. Publicly described data are preclinical, including mouse tumor models in which the candidate reportedly showed stronger tumor growth inhibition than avelumab while maintaining low circulating levels of released warhead.
