AVA7100 is a first-in-class, third-generation, FAP-enabled Affimer drug conjugate (AffDC) developed by Avacta for the treatment of solid tumors. It utilizes Avacta’s proprietary pre|CISION technology to selectively deliver highly potent cytotoxic payloads directly to the tumor microenvironment (TME), minimizing exposure in normal tissues. The drug consists of a novel fibroblast activation protein (FAP)-binding Affimer molecule conjugated via a pre|CISION peptide linker to an undisclosed cytotoxic warhead. This design allows for targeted release of the payload in FAP-expressing tumor stroma, including cancers with low levels of FAP expression. The Affimer scaffold offers advantages over traditional antibodies, such as smaller size and higher binding affinity. As of June 2025, AVA7100 remains in preclinical development with candidate selection anticipated in late 2025 and IND-enabling studies expected thereafter[1][2][3][4][5][7][8][10].
