Avadomide + rituximab + cyclophosphamide + doxorubicin + vincristine + prednisone is an investigational combination regimen under evaluation for the first-line treatment of diffuse large B-cell lymphoma (DLBCL) with poor risk factors[7]. It integrates avadomide, a novel cereblon E3 ligase modulator (CELMoD) with immunomodulatory and antineoplastic activity, into the established R-CHOP regimen. R-CHOP combines rituximab, a chimeric monoclonal antibody targeting CD20 on B-cells; cyclophosphamide, an alkylating agent; doxorubicin, an anthracycline antibiotic; vincristine, a microtubule inhibitor; and prednisone, a corticosteroid. Avadomide induces degradation of transcription factors IKZF1 and IKZF3 via cereblon binding, modulating immune function and promoting antitumor effects. Each component has a distinct but complementary mechanism, leading to synergistic cytotoxicity and immunomodulation for aggressive B-cell malignancies[2][3][7].
