Overview
Avadomide + dexamethasone is a combination regimen consisting of avadomide, an oral small molecule cereblon E3 ligase modulator (CELMoD), and dexamethasone, a synthetic glucocorticoid corticosteroid. **Avadomide** exerts antitumor and immunomodulatory effects by modulating cereblon-dependent ubiquitination, leading to degradation of transcription factors such as Ikaros and Aiolos, resulting in antiproliferative and pro-apoptotic activity. It is under investigation for hematologic malignancies, including non-Hodgkin lymphoma and multiple myeloma, and has dose-limiting toxicities such as neutropenia[3]. **Dexamethasone** enhances antitumor activity by anti-inflammatory and immunosuppressive actions through the glucocorticoid receptor. The combination aims to optimize efficacy via synergistic immune-modulatory and antiproliferative effects.
