CC-122 + CC-292 is an investigational combination therapy consisting of the cereblon E3 ligase modulator (CELMoD) **avadomide** (CC-122) and the Bruton's tyrosine kinase (BTK) inhibitor **spebrutinib** (CC-292). Developed by Celgene (now part of Bristol Myers Squibb), the combination was designed to synergistically target B-cell malignancies by promoting the degradation of transcription factors Aiolos and Ikaros while simultaneously inhibiting B-cell receptor signaling. It was evaluated in a Phase Ib clinical trial (NCT02031419) for relapsed/refractory diffuse large B-cell lymphoma (DLBCL) and follicular lymphoma. However, the combination was associated with enhanced toxicity and insufficient improvement in clinical responses compared to other regimens, leading to the termination of its development for these indications.
