avatrombopag + fostamatinib

Avatrombopag + fostamatinib is a combination therapy used primarily for the treatment of multirefractory immune thrombocytopenia (ITP). This combination targets different pathophysiological pathways in ITP. Avatrombopag is a thrombopoietin receptor (TPO-R) agonist that stimulates platelet production by promoting proliferation and differentiation of megakaryocytes from bone marrow progenitor cells. Fostamatinib is a spleen tyrosine kinase (SYK) inhibitor that blocks the cascade of signals mediated by SYK after the activation of FcγRs, reducing macrophage activity, B lymphocyte proliferation, and antibody production. This dual approach addresses both impaired platelet production and increased platelet destruction in ITP patients.
