AVB-406 is a high-affinity AXL decoy receptor designed to inhibit the GAS6/AXL signaling axis, a pathway frequently implicated in tumor progression, metastasis, and resistance to therapy. Developed by Aravive Biologics (now Aravive), AVB-406 is an engineered version of the extracellular domain of the AXL receptor that binds to its ligand, Growth Arrest-Specific 6 (GAS6), with an affinity several orders of magnitude higher than the wild-type receptor. By acting as a "GAS6 trap," AVB-406 effectively sequesters the ligand and prevents it from activating endogenous AXL receptors on the surface of cancer cells. This inhibition suppresses downstream signaling pathways involved in epithelial-to-mesenchymal transition (EMT) and cell survival. While AVB-406 showed significant promise in preclinical models of ovarian, breast, and pancreatic cancers, Aravive's clinical development program prioritized AVB-S6-001 (batiraxcept), a related Fc-fusion protein with optimized pharmacokinetic properties.
