AVE0847

AVE0847 is an orally active small molecule that acts as a dual inhibitor of the sodium-hydrogen exchanger 3 (NHE3) and the sodium-glucose cotransporter 1 (SGLT1). Developed by Sanofi, it was primarily investigated for the treatment of type 2 diabetes mellitus. The mechanism of action involves the inhibition of SGLT1 in the gastrointestinal tract, which delays and reduces the absorption of dietary glucose, thereby lowering postprandial glycemic excursions. Simultaneously, the inhibition of NHE3 in the intestine reduces sodium absorption, which may provide additional therapeutic benefits in blood pressure regulation and fluid balance. Although the drug reached Phase 2 clinical trials, its development was ultimately terminated by Sanofi.
