Overview

AVE1642 + sorafenib is a combination of a **humanized monoclonal antibody (AVE1642) targeting type 1 insulin-like growth factor receptor (IGF-1R)** and **sorafenib**, a small-molecule multikinase inhibitor. AVE1642 blocks IGF-1R signaling, which is implicated in tumor cell growth and resistance to therapy. Sorafenib inhibits several receptor tyrosine kinases, including VEGFR, PDGFR, and RAF kinases, and is approved for the treatment of hepatocellular carcinoma and renal cell carcinoma. The combination has been investigated in early phase clinical trials for patients with advanced hepatocellular carcinoma and other solid tumors, aiming to explore safety, tolerability, pharmacokinetics, and preliminary efficacy. AVE1642 is administered intravenously, while sorafenib is given orally. Most patients experienced tumor stabilization, and no significant pharmacokinetic interactions or increased toxicities were observed when combining both agents[2].
