AVE9633 is an investigational antibody-drug conjugate (ADC) composed of a humanized monoclonal antibody targeting the myeloid cell surface antigen CD33 linked to the cytotoxic maytansinoid DM4. The monoclonal antibody portion specifically binds to CD33 expressed on myeloid leukemia cells; after internalization, the DM4 moiety is released inside the cell where it binds tubulin and disrupts microtubule assembly/disassembly dynamics. This results in mitotic arrest and apoptosis of targeted cells. AVE9633 was developed for the treatment of acute myeloid leukemia (AML), particularly in patients with relapsed or refractory disease. It was created by ImmunoGen and further developed in collaboration with Sanofi. Phase I clinical trials were conducted but discontinued early due to limited efficacy at doses above those required for CD33 saturation[1][2][3][5].
