Avelumab is a fully human monoclonal antibody that targets programmed death-ligand 1 (PD-L1), functioning as an immune checkpoint inhibitor. By binding to PD-L1, it blocks its interaction with the inhibitory T-cell receptor PD-1 and B7.1, thereby removing suppression of T-cell activity and promoting adaptive antitumor immune responses. Unlike some other anti-PD-L1 antibodies, avelumab has a wild-type IgG1 Fc region, enabling engagement with Fc-γ receptors on natural killer cells and inducing antibody-dependent cell-mediated cytotoxicity (ADCC). Avelumab is approved for the treatment of metastatic Merkel cell carcinoma (MCC), metastatic urothelial carcinoma (UC), and advanced renal cell carcinoma (RCC) in combination with axitinib. It was developed by Merck KGaA and Pfizer[3][5][6].
